I always loved the scene between Wiz and Travis in Taxi Driver where they go outside and Travis tries to get some advice on feeling 'real down'. It's not so much what Wiz says as the obvious sincerity with which he says it - under normal circumstances, it would probably help. Unfortunately, Travis is too far gone for it to do any good. A beautiful, sad scene, played to absolute perfection by De Niro and Peter Boyle.
Edit: I now see the specification 'TV characters'. You don't mind if I ignore that, do you?

I've always liked the friendship between Saga and Martin on the Scandinavian crime drama, Bron/Broen. It started as the classic 'mismatched cops' formula, but throughout the two seasons it blossomed into something really beautiful and complex. Both clearly respect each other and while they don't always understand the other person's boundaries, there is a deep undercurrent of care between the two. There's one scene at the end of the first scene (no, I won't spoil it) where Saga lies to Martin to prevent him from doing something extreme and knowing her reasons why make the moment all the more sad.
On the romance front, Violet and Sian's relationship on Banana was so sweet and yet still acknowledged that because both women were young, there was no guarantee their love would last, even if they genuinely liked one another. I really enjoyed that.

Buffy/Willow and Buffy/Giles. Icabod/Abbie on Sleepy Hollow. Mulder/Scully, in the non-stupid seasons of The X-Files. Zoe/Wash, River/Book, Mal/Kaylee on Firefly. Shawn/Gus on Psych. Amantha/Daniel and Daniel/Tawney on Rectify. Todd/Mr. Peanutbutter, the only even slightly healthy relationship on BoJack Horseman! Finn and Jake, of course. Mabel/Waddles on Gravity Falls. Chuck/Morgan on Chuck.

Oh god, the reveal in "Standing" from Once More With Feeling floored me and I still get a little emotional thinking about Giles taking off on her. Anthony Stewart Head's awesome vocals really sold it.
And it doesn't really count, but I love the relationship between Doc Hammer and Jackson Publick. I just love listening to their idiotic banter on the Venture Bros DVDs while hanging out in Astro-Base Go. I guess it reminds me of an earlier life shooting the shit with my roommate telling stupid jokes.

Abby & Ichabod on Sleepy Hollow! A rare depiction of interracial/cross-gender platonic friendship that is totally believable and also very fun. They have chemistry, but it's not at all forced. I just hope they don't get together in some sort of ratings ploy (though I wouldn't mind seeing either of those leads naked - they are both beautiful).

Shawn/Gus on Psych JD/Turk on Scrubs I thought those two were incredibly funny.
Bashir and O'Brien on ST: Deep Space Nine. Best friendship in Star Trek.
I also really loved the relationship between Carter and Benton on ER. That was a nice depiction of a workplace relationship.
Locke and Ben on Lost, which was never boring.
Lorelai and Rory on Gilmore Girls Rory and Paris on Gilmore Girls This show really had some wonderfully developed relationships
Frasier and Niles, on Frasier. This was a great depiction of siblings.

Pam/Michael on The Office-
I'm a huge Office fan and oh man that relationship. I can sum it all up in the scene: when she has her art show and he shows up at the end and is impressed. And she spontaneously hugs him when he says he's proud of her. She had just been waiting for that ALL night. For someone to make her feel proud of what she'd done. I can tear up right freaking now.
Leslie/Ann on P&R-
A fucking refreshing, honest look at a female friendship which is so damn real it makes me want to go hug my besties.
April/Andy on P&R-
I deserve a punch in the face on this one, or at the very least an eye-roll. But they remind me so much of me and my boyfriend. Some of the character similarities are ridiculous, but I'll just say that if we lived together we'd blow all our money on video games or toys or something silly, and be more than happy to eat chili out of a frisbee cause we had no plates. I love the childlike nature of their relationship.

I have a deep affection for Margot & Alana's relationship on Hannibal. They've both been through so much shit it's nice to see them happy and victorious for once, y'know? It helps that they're just really cute together.
Jack and Bella deserve a mention as well. For a character who everyone knew was doomed by the end of her first episode, watching Bella's gradual decline hurt. Her funeral in "Aperitivo" is crushing, and her loss is felt through the rest of the series- Jack basically loses touch with his humanity after that point.
(Will and Hannibal were the obvious pick here, but their thing is pretty horrifying on most levels...)
Most of the others I was gonna pick were covered by Mrs Langdon Alger!

Jack Donaghy & Liz Lemon: I love how Jack and Liz’s friendship evolved from episode to episode, from season to season. While Jack is often portrayed as a selfish capitalist who’s concerned about how he’s going to get to where he wants to be, there’s a selflessness in his relationship with Liz, even with his constant poking fun at her and downplaying her problems, that not only keeps it together, but strengthens it. And it’s always nice when Liz is able to return the favor or prove that she’s more formidable or experienced in a certain way – in any way – than Jack expected. One of my favorite “30 Rock” moments is when Jack abandons GE for a short-lived stint at the Bush administration where Liz is back at New York going through a (false) pregnancy scare, and she keeps calling Jack for advice and help but he’s unable to get back to her. At the end of the episode, he says to her, “I’m sorry you had to go through this alone.” It’s a really poignant moment that sums up the friendship well, and it’s something I don’t think too many people would be able to say to a friend they’ve let down or forgotten about, let alone someone as powerful, determined and busy as Jack.
Michael Scott & Dwight Schrute: I’ve said before that Michael treats Dwight as if he is the nerdy younger brother to the coolest kid in school who shows up at inconvenient times to mess with his otherwise sterling reputation. Obviously, with Michael’s perception of himself that’s true, but when you consider his employees’ perspective of him, it’s not. The half-disgust, half-serious validation Michael gets from Dwight’s nerdy, loyal suck-up is defining, and it’s always nice when they have moments of real connection that Michael doesn’t try to deny.
Homer Simpson & Ned Flanders: Nothing more needs to be said.
